چکیده

BACKGROUND: Bone Morphogenetic Protein Receptor II (BMPR2) deficiency is associated with the pathologic development of pulmonary vascular changes in Pulmonary Arterial Hypertension (PAH). Fibroblast most abundant cell vascular. However, there only a little information regarding effect BMPR2 fibroblast. This study aims to understand fibroblasts.METHODS: applied CRISPR/Cas9 technique edit BMPPR2 NIH-3T3 cells. The transfection for editing into cells was done by using chitosan nanoparticles. evaluation and Transforming Growth Factor (TGF)-? mRNA expression Quantitative real-time polymerase chain reaction. assessment edited proliferation scratch test assay.RESULTS: CRISPR/Cas9-edited group lower than untreated group. higher TGF-? groups similar.CONCLUSION: fibroblast increase ability proliferate.KEYWORDS: BMPR2, PAH, NIH-3T3, CRISPR/Cas9,

Diffusion capacity and BMPR2 mutations in pulmonary arterial hypertension.

Self-reported risks for multiple-drug resistance among new tuberculosis cases: Implications for drug susceptibility screening and treatment. Pulmonary arterial hypertension (PAH) is a disease in which remodelling of the small pulmonary arteries leads to an increase in pulmonary artery pressure (PAP). The most important genetic predisposing factor related to PAH is a mutation in the bone morphog...

Increased susceptibility to pulmonary hypertension in heterozygous BMPR2-mutant mice.

BACKGROUND Bone morphogenetic protein receptor-2 (BMPR2)-heterozygous, mutant (BMPR2(+/-)) mice have a genetic trait similar to that of certain patients with idiopathic pulmonary arterial hypertension (IPAH). To understand the role of BMPR2 in the development of IPAH, we examined the phenotype of BMPR2(+/-) mice and their response to inflammatory stress. METHODS AND RESULTS BMPR2(+/-) mice we...

BMPR2 germline mutations in pulmonary hypertension associated with fenfluramine derivatives.

This study investigated whether patients developing pulmonary arterial hypertension (PAH) after exposure to the appetite suppressants fenfluramine and dexfenfluramine have mutations in the bone morphogenetic protein receptor 2 (BMPR2) gene, as reported in primary pulmonary hypertension. BMPR2 was examined for mutations in 33 unrelated patients with sporadic PAH, and in two sisters with PAH, all...

Relationship of BMPR2 mutations to vasoreactivity in pulmonary arterial hypertension.

BACKGROUND Vasoreactivity tests are fundamental in evaluating pulmonary arterial hypertension (PAH). Mutations of the transforming growth factor-beta type II receptor gene, BMPR2, predispose to the development of pulmonary hypertension and may alter the response to vasodilators. Previous investigations have not examined the relationship of BMPR2 mutations to vasoreactivity. METHODS AND RESULT...

Targeted gene delivery of BMPR2 attenuates pulmonary hypertension.

Pulmonary arterial hypertension (PAH) remains a fatal disease despite modern pharmacotherapy. Mutations in the gene for bone morphogenetic protein receptor type II (BMPR2) lead to reduced BMPR2 expression, which is causally linked to PAH. BMPR2 is predominantly expressed on pulmonary endothelium and has complex interactions with transforming growth factor (TGF)-β signalling mechanisms. Our obje...
